Product Overview

Biochemazone 0.1 M Phosphate Buffer, pH 7.4 BZ204 is a sterile sodium phosphate buffer made from disodium hydrogen phosphate and sodium dihydrogen phosphate. It holds a near-physiological pH 7.4 at 25 °C and contains no sodium chloride, so it is ideal where a plain phosphate buffer – not PBS – is called for.

Phosphate buffers work best around pH 5.8–8.0, centred on the second pKa of phosphoric acid (about 7.2). The 0.1 M strength is the classic concentration for perfusion and fixative preparation (e.g. 4% paraformaldehyde in 0.1 M PB), tissue rinsing, enzyme assays and general dilution. The buffer is sterile-filtered through a 0.2 µm membrane.

Each batch is prepared with ISO 3696 compliant high-purity water under Biochemazone’s ISO 9001 / GMP quality system.

Frequently asked questions

What is the difference between phosphate buffer (PB) and PBS?

PB contains only sodium phosphate salts. PBS also contains sodium chloride and potassium chloride to make it isotonic. Many fixation and perfusion protocols ask for plain 0.1 M PB.

Can I make 4% PFA with this buffer?

Yes. 0.1 M PB pH 7.4 is the standard base for 4% paraformaldehyde fixative. Dissolve the PFA according to your protocol and cool before use.

The buffer has crystals after cold storage. Is it OK?

Yes. Phosphate salts can crystallise in the cold. Warm to room temperature or 37 °C and mix until fully clear.

Can I dilute it to 0.05 M or 0.01 M?

Yes. Dilute with high-purity water. The pH changes only slightly on dilution; check it if your method needs an exact value.

How should 0.1 M phosphate buffer be stored?

Store at 2–30 °C, tightly closed. Unopened bottles have a 1-year shelf life and ship at room temperature.
