Biochemazone Artificial Lysosomal Fluid BZ257, also called ALF or simulated lysosomal fluid, is a sterile, ready-to-use solution that simulates the composition and pH of lysosomal fluid, the acidic fluid inside lysosomes. It can be used directly and is supplied at pH 4.5; the stabilizer, pH and contents can be customized.
Lysosomes are acidic compartments of the cell, filled with more than 60 different types of hydrolases. They mediate the degradation of extracellular particles taken up by endocytosis and of intracellular components from autophagy; the digested products leave the lysosome through specific catabolite exporters or by vesicular membrane trafficking. BZ257 gives laboratories this acidic environment as a ready-made fluid, for in vitro dissolution and biodurability studies.
It is supplied stabilized and has a 1-year shelf life unopened; a preservative-free version (6-month shelf life) and a dried Powder Version Kit are available on request. The stabilizer, pH and contents can be customized to the project. This simulated fluid is for scientific research and in vitro use only; it is not for clinical diagnosis or treatment. How does ALF differ from Gamble’s solution?
ALF simulates the acidic fluid inside lysosomes, at pH 4.5. Gamble’s solution represents the interstitial fluid deep within the lungs, at pH 7.4; see Gamble’s Solution Lung Fluid (BZ284).
How long does the solution keep?
Unopened, the stabilized solution has a 1-year shelf life at 2–30 °C. A preservative-free version with a 6-month shelf life and a dried Powder Version Kit with a 2-year shelf life are available on request.
