Product Overview

Biochemazone Sodium Citrate Buffer Powder BZ213 is a pre-weighed dry blend that makes 10 mM sodium citrate buffer, pH 6.0 – the most widely used antigen retrieval buffer for immunohistochemistry (IHC) and immunocytochemistry. Simply dissolve each pack in deionised or distilled water to the stated volume.

Formaldehyde fixation forms cross-links that mask many epitopes. Heating sections in citrate buffer pH 6.0 (heat-induced epitope retrieval, HIER) reverses much of this masking and restores antibody binding on formalin-fixed paraffin-embedded (FFPE) and frozen sections. As a guide, 1 L of working buffer covers about 100 slides at 10 mL per slide. The dry format ships light, stores for 2 years and avoids weighing and pH adjustment.

Each batch is blended and packed under Biochemazone’s ISO 9001 / GMP quality system.

Composition (per litre of prepared buffer)

ComponentConcentrationAmount per litre
Trisodium citrate dihydrate≈8.1 mM≈2.4 g
Citric acid monohydrate≈1.9 mM≈0.4 g
Total citrate10 mM—
Deionised or distilled water (added by user)—to 1 L

Gives pH 6.0 at 25 °C when dissolved to the stated volume. Optional: add 0.05% Tween 20 (0.5 mL per litre) if your protocol calls for it.

How to Use (HIER Guide)

StepWhat to do
1. Prepare bufferDissolve one pack in about 90% of the final volume of water, then make up to the stated volume (1, 2, 5 or 10 L).
2. Dewax and rehydrateDewax FFPE sections in xylene and rehydrate through graded ethanol to water.
3. HeatHeat slides in the buffer at 95–100 °C for 10–20 min (water bath, steamer, microwave or pressure cooker).
4. CoolLeave slides in the buffer to cool for about 20 min at room temperature.
5. Continue IHCRinse in PBS or TBS wash buffer and continue with blocking and antibody steps.

Optimise heating time and temperature for each antibody and tissue.

Frequently asked questions

How do I prepare the buffer?

Dissolve one pack in about 90% of the final volume of deionised or distilled water, mix until clear, then make up to the volume stated on the pack. No pH adjustment is needed.

How many slides does 1 L cover?

At about 10 mL per slide, 1 L of working buffer covers roughly 100 slides. Coplin jars and staining racks use different volumes.

Should I add Tween 20?

It is optional. Some protocols add 0.05% Tween 20 to improve wetting; follow your antibody datasheet.

Citrate pH 6.0 or Tris-EDTA pH 9.0?

Citrate pH 6.0 suits most antibodies and preserves tissue morphology well. Some epitopes retrieve better at pH 9.0; check the antibody datasheet.

How should the powder be stored?

Store sealed packs dry at 2–30 °C. Unopened packs have a 2-year shelf life and ship at room temperature.
