How do I make 1X TBST from BZ226?
Mix 100 mL of BZ226 with 900 mL of high-purity water. The 1X buffer contains 50 mM Tris-HCl, 150 mM NaCl and 0.1% Tween 20 at pH 7.4.
Should I use TBST or PBST for Western blots?
Both work for most antibodies. TBST is preferred for phospho-specific antibodies and alkaline phosphatase detection because phosphate can interfere. PBST (BZ219) is a common choice for general and ELISA washing.
Can I make blocking buffer with TBST?
Yes. Dilute to 1X, then add 5% non-fat dry milk or 3–5% BSA to make blocking buffer or antibody diluent.
How many washes are recommended?
A typical protocol uses three washes of 5–10 minutes with 1X TBST after each antibody incubation, with gentle shaking.
How should 10X TBST be stored?
Store at 2–30 °C with the cap tightly closed. Unopened bottles have a 1-year shelf life and ship at room temperature.
