Product Overview

Biochemazone 50X TAE Buffer BZ208 is a fifty-times concentrated Tris-acetate-EDTA buffer for agarose gel electrophoresis of DNA and RNA. Diluted to 1X it is used to cast the gel and as the running buffer, and one litre of concentrate makes 50 litres of working buffer.

TAE gives fast migration of linear double-stranded DNA and good separation of large fragments, genomic DNA and supercoiled plasmids. Because it contains no borate, it is the preferred buffer when DNA is to be recovered from the gel for ligation, cloning or sequencing. The concentrate is filtered through a 0.22 µm membrane.

TAE has a lower buffering capacity than TBE, so replace or recirculate the buffer during long runs. Each batch is prepared with ISO 3696 compliant high-purity water under Biochemazone’s ISO 9001 / GMP quality system.

Frequently asked questions

How do I dilute 50X TAE to 1X?

Add 20 mL of BZ208 to 980 mL of high-purity water (1 part + 49 parts). Use 1X TAE both to cast the agarose gel and to fill the electrophoresis tank.

When should I choose TAE instead of TBE?

Choose TAE for large DNA fragments, genomic DNA and supercoiled plasmids, and whenever DNA will be recovered from the gel for enzymatic steps. Choose TBE (BZ202) for small fragments, PAGE and long high-voltage runs.

Why does TAE need replacing during long runs?

TAE has a lower buffering capacity than TBE. During long or high-voltage runs the buffer becomes exhausted, so replace it or recirculate it between the electrodes.

Is BZ208 filtered?

Yes. The concentrate is filtered through a 0.22 µm membrane.

How should 50X TAE be stored?

Store at 2–30 °C, tightly closed. Unopened bottles have a 1-year shelf life and ship at room temperature.
