Frequently asked questions
How do I make 1X or 0.5X TBE from BZ202?
For 1X, mix 1 part BZ202 with 9 parts high-purity water. For 0.5X, the most common agarose running strength, mix 1 part with 19 parts water. Use the same dilution for the gel and the running buffer.
Should I use TBE or TAE?
Use TBE for small DNA fragments, PAGE and long runs, because it buffers more strongly and gives sharper bands. Use TAE (BZ208) for large fragments and when DNA will be cut from the gel for cloning, because borate inhibits many enzymes.
There is a white precipitate in the bottle. What should I do?
Concentrated TBE can precipitate in the cold. Warm to 37 °C and mix until clear before diluting. If a heavy precipitate does not dissolve, do not use the buffer.
What is the pH of the TBE buffer?
After dilution to 1X the pH is about 8.3, the standard value for Tris-borate-EDTA running buffer.
How should 10X TBE be stored?
Store at 2–30 °C, tightly closed. Unopened bottles have a 1-year shelf life and are shipped at room temperature.
